In order to unlock the shelters you will have to complete a mission. This mission is called Home Expansion and to access it you will have to go to any of the train stations around the map.

At each station there will be a notice board with different quests you can get. Interact with the Vaults sign on one of the boards to start the home expansion quest, which will allow you to access the Vault 51 Vaults Claim Center.

To get to Vault 51 you will have to go to the northwest corner of the map, high up in the mountains, and upon arrival, you will have to interact with a Mr. Clark robot. To complete the quest you will need to register as a shelter owner, which will allow you to build a basic shelter.
