The first one is by finding a U-bend in your loop and filling the surrounding spaces using specific buildings. Then you will have to fill in the spaces using Spider Cocoons. Inside the U-turn, for which you will need a vampire mansion.

This will give you the certainty that each Spider Cocoon has at least four enemies per loop. You could start with a single Spider Cocoon and build as you get gear.

Another way is to place a Grove on either side of a Chronos Crystal, which will produce twice the wolf rat, increasing your chances of getting even more than four enemies per token. The Rat Wolves move between tiles, so you will have less control, although at least there will be no vampires here, but you will still have to add one to a U-curve to increase the chances.
